Pharmacological action
Afoxolaner blocks the transmission of chloride ions from the presynaptic membrane to the postsynaptic membrane by acting on ligand-gated chloride ion channels, especially inhibiting channels gated by the neurotransmitter γ-aminobutyric acid (GABA), resulting in increased activity of insect neurons and death due to excessive excitement.
This mechanism enables Afoxolaner to effectively kill fleas and ticks.
Clinical Applications
Afoxolaner is mainly used to treat flea and tick infestations in dogs. Specific applications include:
Flea infestation: kill fleas within 8 hours.
Tick infestation: kill ticks within 48 hours.
Mite infestation: treat canine mite infestations (such as canine scabies and canine demodex).
Dosage and Usage
Afoxolaner is usually used in the form of chewable tablets, and the specific usage is as follows:
Dosage for dogs: Administer according to body weight, with a dose range of 2.7mg/kg to 7.0mg/kg. Administer once a month, adjusted according to epidemiological conditions.
Treatment of scabies infestation: Administer once a month, twice in a row, and the administration time can be extended if necessary
