Madecassoside is a major bioactive saponin compound derived from the herb Centella asiatica (L.) Urb., which is commonly found in tropical and subtropical regions of Asia. It is one of the primary triterpenoid constituents of this traditional medicinal plant, known for its various biological activities and therapeutic potentials.
Madecassoside has been reported to possess anti-inflammatory, antioxidant, neuroprotective, and cardioprotective effects. It has been used in traditional medicine for the treatment of conditions such as traumatic injuries, keloids, and scars. The compound has also shown promise in wound healing, with studies suggesting it can promote skin angiogenesis and collagen synthesis, as well as exhibit antifibrotic properties.
Pharmacokinetic studies have shown that madecassoside is widely distributed in the heart, liver, spleen, lungs, and kidneys of rats after oral administration. It is excreted primarily through bile, urine, and feces, with the majority of the compound being eliminated in the feces as metabolites. The compound has been found to interact with hepatobiliary efflux transporters, suggesting that P-glycoprotein and multidrug resistance-associated protein contribute to its hepatobiliary elimination.
Madecassoside has a molecular formula of C48H78O20 and a molecular weight of 955.14 g/mol. It is known to have a significant impact on the production of pro-inflammatory cytokines and has been shown to inhibit the nuclear translocation of NF-κB in Propionibacterium acnes-stimulated THP-1 human monocytic cells. Additionally, it has been demonstrated to enhance skin hydration by increasing the expression of aquaporin-3, loricrin, and involucrin in HaCaT keratinocytes, as well as promoting hyaluronan secretion in human dermal fibroblasts.
