Rapamycin is a white solid crystal with a melting point of 183-185°C. It is lipophilic and soluble in organic solvents , ethanol, . It is very slightly soluble in water and almost insoluble in ether. Clinically, it is a new macrolide immunosuppressant. Rapamycin blocks signal transduction through different cytokine receptors, blocking the progression of T lymphocytes and other cells from the G1 phase to the S phase, thereby exerting an immunosuppressive effect.
English aliases: RPM;RAPA;sirolimus(Sirolimus);RAPAMUNE;Rapamycin/Sirolimus;Rapamycin(Sirolimus);Rapamycin;RAPAMYCIN 1GM;SIROLIMUS;23,27-Epoxy-3H-pyrido[2,1-c][1,4]oxaazacyclohentriacontine,AY 22989,Sirolimus;sila9268a;Rapamycin from Streptomyces hygroscopicus;RAPAMYCIN;
Synthesis route
Rapamycin is synthesized from seven units of acetate and seven units of propionate through the polyketide pathway, and the required O-methyl comes from methionine. In fact, the nitrogen source is a derivative of shikimic acid after reduction, Lysine is first deaminated to form carboxylic acid piperidine, which is then connected to the polyketide acetyl bond and amide bond by carboxylic acid piperidine to form the initial structure of rapamycin.

Uses
A new and highly effective immunosuppressant, clinically used for the treatment of organ transplant rejection and autoimmune diseases.
The latest research from the University of Texas in the United States shows that rapamycin can be used to treat Alzheimer's disease. The drug ingredient is also found in the secretions of bacteria in the soil of Easter Island.
In March 2010, according to foreign media reports, scientists recently discovered another use for it: it can be used to treat Alzheimer's disease (senile dementia).
