Description


β-Nicotinamide Mononucleotide is also known as nicotinamide mononucleotide. Its chemical formula is C11H15N2O8P. It is easily soluble in water and is a white to slightly yellow crystalline powder with no obvious odor. It needs to be kept away from light and waterproof at room temperature (the shelf life is 24 months). It has weak acidity (pH = 3~4) and is widely used in drugs, medical and cosmetic raw materials and food additives. The industrial synthesis methods include chemical method and biological method.
NMN is a naturally occurring and biologically active nucleotide that is widely present in the human body, vegetables, fruits and meat. It is particularly abundant in avocados and edamame. It is not only a precursor for the synthesis of coenzyme I, but also an important metabolic intermediate in cells [1]. It belongs to the category of vitamin B derivatives (nicotinamide belongs to vitamin B3). It is closely related to many biochemical reactions such as human immunity, metabolism, and exercise. It has the effects of reducing fat and weight, delaying aging, and treating cardiovascular diseases. Therefore, it has received widespread attention in recent years.

At present, the methods for exogenous preparation of NMN are mainly chemical synthesis and biological methods. The former was studied earlier and mainly uses nicotinamide, ribose and other substances as raw materials through different synthesis steps. However, the complex technical route, harsh reaction conditions, high production cost, low product yield and purity, etc. have greatly limited the application and promotion of the product; however, compared with the chemical synthesis method, the biological method has the advantages of simple technical route, convenient operation, green environmental protection and low production cost.
 

Application & Funtion

 

NMN is an endogenous substance that can be synthesized in vivo by the following reactions. On the one hand, 1 molecule of nicotinamide and 1 molecule of 5-phosphoribosyl-1-pyrophosphate (PRPP) can generate 1 molecule of NMN and 1 molecule of pyrophosphate (PPi) under the catalysis of nicotinamide phosphoribosyltransferase (NAMPT or NAMPRT); on the other hand, 1 molecule of nicotinamide riboside (NR) can also generate 1 molecule of β-nicotinamide mononucleotide under the catalysis of nicotinamide riboside kinase (NRK).

The advantages of using bioenzymatic method to synthesize NMN are that it does not produce harmful substances, is green and environmentally friendly, (bioenzyme) catalysts can be reused, costs are reduced, and conditions are mild.
This method uses nicotinamide, pyrophosphate, adenine nucleotides, etc. as raw materials. Under the catalysis of adenine phosphoribosyl transferase (APRT) and nicotinamide phosphoribosyl transferase (Nampt), the reaction can occur at 30~50 ºC and pH = 6.5~8.5, and NMN can be synthesized.
